Abstract Background (−)-Limonene, one of cyclic monoterpenes, is an important renewable compound used widely as a key building block for the synthesis new biologically active molecules and fine chemicals. (−)-Perillamine, derived from (−)-limonene, highly useful synthon constructing more complicated functionally relevant Aim We aimed to report sustainable efficient method regiospecific C–H amination (−)-limonene into (−)-perillamine. Results Here, we artificial penta-enzymatic cascade system transformation cheap easily available (−)-perillamine first time. This composed cytochrome P450 monooxygenase, alcohol dehydrogenase w-transaminase main reactions, well formate NADH oxidase cofactor recycling. After optimization multi-enzymatic system, 10 mM was smoothly converted 5.4 in one-pot two-step biotransformation, indicating feasibility C7-regiospecific inert bond (−)-limonene. represents concise route biocatalytic derivatives similar natural products.
